High-resolution melting (HRM) analysis has been improved and applied for the first time to quantitative analysis of enzymatic reactions. By using the relative ratios of peak intensities of substrates and products, the quantitativity of conventional HRM analysis has been improved to allow detailed kinetic analysis. As an example, the ligation of sticky ends through the action of T4 DNA ligase has been kinetically analyzed, with comprehensive data on substrate specificity and other properties having been obtained. For the first time, the kinetic parameters (kobs and apparent Km) of sticky-end ligation were obtained for both fully matched and mismatched sticky ends. The effect of ATP concentration on sticky-end ligation was also investigated. The improved HRM method should also be applicable to versatile DNA-transforming enzymes, because the only requirement is that the products have Tm values different enough from the substrates.  相似文献

In this work, we investigate the influence mechanism of hydrogen partial pressure on fracture toughness and fatigue life of a high strength pipeline steel. Both fracture toughness test and fatigue life test are carried out under different hydrogen partial pressure. The experimental results show that with the increasing of hydrogen partial pressure, fracture toughness and fatigue life decrease and the decrease trends gradually flatten out. Hydrogen has a larger effect on fatigue life than fracture toughness. Only 3% hydrogen gas can cause a 67.7% decrease of fatigue life. The in-situ hydrogen permeation test is performed respectively in 2 MPa, 5 MPa and 8 MPa hydrogen partial pressure. With the increasing of hydrogen partial pressure, the increase trend of hydrogen permeation current gradually tends to be gentle, which indicates that the hydrogen atoms entering into the material gradually become saturated. This result can be used to clarify the influence mechanism of hydrogen partial pressure on fracture toughness and fatigue life.  相似文献

The use of low‐rank coal in a clean and efficient manner is a major challenge facing the current coal technology. A high‐sulfur coal with 4.5 wt% sulfur is chosen to examine the compatibility of the pristine coal and the purified contrast with a solid oxide fuel cell (SOFC) with nickel cermet anodes. Desulfurization of the pristine coal is performed by molten caustic leaching method with a removal ratio of 80%. Analyses of the physicochemical properties of coal samples indicate that the purified coal has a more favorable structure and higher Boudouard reactivity, which is suitable as a fuel for fuel cells. The assessment of electrochemical performance reveals that the purification treatment not only makes the peak power density of SOFCs improve from 115 to 221 mW cm?2 at 900°C but also extends their durability from 1.7 to 11.2 hours under a current density of 50 mA cm?2 at 850°C with a fuel availability increasing from 6.25% to 40%. The postmortem analyses show that far less deposited carbon and nickel sulfide are observed on the anode surface. The fuel‐based investigation reveals that the purified coal is a promising fuel for direct carbon fuel cells.  相似文献

Experimental methods and theoretical analysis are employed to investigate effects of channel opening area on downward flame spread characteristics of extruded polystyrene (XPS) thermal insulation materials on building facade. The average flame height first drops and then rises as dimensionless opening area (the ratio of sidewall opening area to sidewall area, ie, S*) increases. As S* rises, both the average and maximum temperature of the curtain wall decrease, and the decreasing of the average temperature is linear. XPS surface temperature history can be divided into four stages, ie, inapparent rising stage (preheating), significant rising stage (melting), dropping stage (pyrolysis), and rerising stage (ignition). The preheating length first rises and then drops as S* increases. The XPS flame spreads steadily at the early period while acceleration occurs at the later period. For different opening areas, the difference in spread distance history is not apparent in the early stage while this difference is significant in the later stage. The flame spread rate (Vf) first increases and then decreases as S* rises. A downward flame spread model for XPS in vertical channel with openings is built. The varied trend of Vf predicted using this model corresponds to the experimental result.  相似文献

TaC ceramics with 0.03–0.60?wt% of boron additions were prepared by hot pressing at 2100?°C for 1?h under a pressure of 40?MPa. Effects of boron content on densification, phase composition, microstructure, mechanical properties and oxidation resistance of the TaC ceramics were investigated. When the boron content was 0.12?wt% and above, full density was obtained due to reactions between boron and oxygen impurity at presence of TaC. Minor phases of TaB2 and C were formed in the 0.24 and 0.60?wt% B compositions after gas-out of the oxygen impurity. Microstructure of the TaC ceramics was refined with increasing in boron content. The TaC ceramic with 0.24?wt% of boron showed the best mechanical properties with a Vickers hardness, flexural strength and fracture toughness of 17.7?GPa, 534?MPa and 4.6?MPa?m1/2, respectively. When more boron was added, interfacial bonding of the TaC grains was strengthened causing a decrease in fracture toughness. Oxidation resistance of the TaC ceramics increased with boron content. Particularly, the 0.60?wt% B composition showed a weight gain of 0.0018?g/cm2 after oxidization at 800?°C in air for 3?h.  相似文献

A series of high-response and fast-response/recovery n-butanol gas sensors was fabricated by adding ZnO to In2O3 in varying molar ratios to form ZnO-In2O3 nanocomposites via a facile co-precipitation hydrothermal method. Morphological characterizations revealed that the shape of pure In2O3 was changed from irregular cubes into irregular nanoparticles, 30–50?nm in size, with the addition of ZnO. Compared with the pure In2O3 gas sensor, the ZnO-In2O3 gas sensor exhibits superior n-butanol sensing performance. With the introduction of ZnO, the response of the sensor to n-butanol was improved from 17 to 99.5 at 180?°C for a [Zn]:[In] molar ratio of 1:1. In addition, the ZnO-In2O3 gas sensors show a reduced optimal working temperature, excellent selectivity to n-butanol, and good repeatability. The response of the ZnO-enhanced In2O3-based sensors showed a strong linear relationship with the n-butanol gas concentration, allowing for the quantitative detection of n-butanol gas.  相似文献
